{"id":10395,"date":"2021-07-20T21:03:08","date_gmt":"2021-07-20T19:03:08","guid":{"rendered":"https:\/\/www.nicox.com\/liberation-doxyde-nitrique\/"},"modified":"2023-06-01T14:11:29","modified_gmt":"2023-06-01T12:11:29","slug":"liberation-doxyde-nitrique","status":"publish","type":"page","link":"https:\/\/www.nicox.com\/fr\/liberation-doxyde-nitrique\/","title":{"rendered":"Lib\u00e9ration d&rsquo;oxyde nitrique"},"content":{"rendered":"\n<h2 class=\"wp-block-heading\">L\u2019expertise de Nicox<\/h2>\n\n\n\n<p>Nicox a concentr\u00e9 ses efforts en mati\u00e8re de recherche sur des pathologies oculaires dans lesquelles l\u2019oxyde nitrique (NO) pourrait jouer un r\u00f4le majeur, notamment la r\u00e9duction de la pression intraoculaire (PIO) dans le glaucome ou la sant\u00e9 r\u00e9tinienne.<\/p>\n\n\n\n<p>Le NO est une petite mol\u00e9cule messager naturelle bien connue qui cible une enzyme intracellulaire, la Guanylate Cyclase soluble (GCs), qui convertit la guanosine triphosphate en un second messager la guanosine monophosphate cyclique (GMPc). La voie de signalisation NO\/GCs joue un r\u00f4le cl\u00e9 dans la r\u00e9gulation de l&rsquo;hom\u00e9ostasie de la PIO et du flux sanguin oculaire. L\u2019augmentation, stimul\u00e9e par le NO, de la concentration en GMPc dans le r\u00e9seau trab\u00e9culaire conduit \u00e0 la s\u00e9questration du calcium intracellulaire, \u00e0 la relaxation de ce r\u00e9seau trab\u00e9culaire et, par consequent \u00e0 l\u2019augmentation du flux d\u2019humeur aqueuse sortant du segment ant\u00e9rieur de l\u2019\u0153il par la voie de drainage primaire ou conventionnelle. Tous les m\u00e9canismes pr\u00e9c\u00e9demment d\u00e9crits pourraient conduire \u00e0 la r\u00e9duction de la PIO et \u00e0 l\u2019am\u00e9lioration de la perfusion oculaire. L\u2019effet du NO sur la r\u00e9duction de la PIO pourrait \u00eatre intensifi\u00e9 et prolong\u00e9 par les inhibiteurs de la phosphodiest\u00e9rase 5 (PDE5) qui inhibent la degradation de la GMPc.<\/p>\n\n\n\n<p>Les enseignements cl\u00e9 de nos activit\u00e9s de recherche sont appliqu\u00e9s \u00e0 des groupements donneurs de NO li\u00e9s \u00e0 d\u2019autres compos\u00e9s de classes th\u00e9rapeutiques, non-analogues de prostaglandine (PGAs), dans le but de renforcer les effets modul\u00e9s par le NO.
